Scanner-based systems for localised photomanipulation
The UGA-42 series comprises a variety of scanning devices for photomanipulation in microscopy and macroscopy applications. All devices allow the positioning of a laser beam virtually anywhere in the field of view of the camera for sequential illumination of points and/ or scanning of regions of interest (ROIs).
UGA-42 Firefly
Small spot with high power density for general photomanipulation applications
The UGA-42 Firefly is a point scanning device for photomanipulation, suitable, in particular, for applications which require high power density and/or small spots. It can be equipped with one or more laser sources coupled via optical fibers or directly, via optics, depending on the type and power of the laser. The spot size depends on the configuration of the system (e.g. microscope model, objective magnification & NA, type of laser connection). Spots close to the diffraction limit can be achieved using single mode fibers or directly coupled lasers. Up to four lasers can be used independently within the same experiment
UGA-42 Geo
Variety of preset spot shapes and sizes for illumination of larger areas without scanning Scanning ROIs with a small spot is associated with a temporal delay of illumination within the corresponding region. For fast processes, like neuronal signaling or free diffusion, scanning can lead to undesired artefacts and loss of signal. To overcome this limitation, in applications where simultaneous illumination throughout an area is crucial, Rapp OptoElectronic has developed the UGA-42 Geo, a unique scanning device in which the laser beam is reshaped to produce spots of different sizes and shapes which are illuminated without scanning.
UGA-42 Caliburn
Small spot size lasers for ablation, microdissection and DNA-damage
The UGA-42 Caliburn is designed to produce localized, controlled damage in biological samples.
Similar to the UGA-42 Firefly, the UGA-42 Caliburn produces small laser spots in the microscope
field of view, but, instead of fiber optically coupled CW lasers, it is equipped with directly coupled
pulsed lasers. Different nanosecond pulse lasers at 355 nm and 266 nm are available, depending on
experimental need.
The UGA-42 Caliburn features motorized laser focus and laser power attenuator.

The UGA‑42 Firefly can be easily combined and upgraded with up to four separate laser ports and/ or with all other modules of the UGA‑42 series such as the UGA‑42 Geo selectable spot shape scanning module and/ or the UGA‑42 Caliburn laser ablation module. This modularity makes the UGA‑42 Firefly the ideal system to start with. Furthermore, UGA‑42 devices can be connected to almost all models of major research microscope brands, giving you the possibility to choose the perfect microscope for your applications and individual needs.
High Flexibility
With the intuitive and easy-to-use SysCon software one can generate a diversity of shapes, sizes, customization and grid pattern options to define a region of interest (ROI). The “Click and Fire” mode allows photomanipulation of e.g. moving objects. Different plugins for the SysCon software are available for direct communication with standard imaging software packages.
Precision and Speed
Due to the real-time controller the scan-mirrors of the UGA‑42 Firefly are extremely precise both spatially and temporarily. Based on the setup in the SysCon sequence manger, the laser beam moves accurately and fast across the sample. Easy hardware synchronization with other devices via TTL ports gives you even more possibilities to setup precise and complex experiments by using auxiliary devices.
UGA-42 GEOU Shape Illumination/ Simultaneous Illumination of Large Areas
The UGA‑42 Geo comes with preset hardware-defined geometries like circles, squares, rectangles and donuts of different size. By reshaping the laser spot, simultaneous illumination of a selected geometry is enabled. It replaces the necessity to scan larger ROIs such as entire cells, and avoids a temporal delay during the illumination.
Modularity and Flexibility
The UGA‑42 Geo includes the UGA-42 Firefly which allows free positioning of the chosen shape within the field of view. This increases the flexibility for your application and system. In addition, the UGA‑42 Geo can be combined with the UGA‑42 Caliburn ablation system. Furthermore, in addition to a single line laser light source, a laser combiner like the LDI laser diode illuminator can be coupled to the UGA‑42 Geo module to provide even more flexibility for your present and future experimental needs.
GA-42 Caliburn Modularity
The UGA‑42 Caliburn system comes with the UGA‑42 Firefly and allows to target the laser beam anywhere in the field of view with high precision. In addition, the Caliburn can be easily upgraded with the UGA‑42 Geo selectable shape scanning device. Up to three additional fiber coupled light sources can be used at the same time with the UGA‑42 Caliburn.
High Flexibility
Like all devices of the UGA‑42 series, the UGA‑42 Caliburn can be connected to almost all models of modern research microscope brands, giving you the possibility to choose the perfect microscope for your applications and individual needs. The optical setup gives you the freedom to attach the device as an add-on module to your microscope without interfering the usage of your existing setup (e.g. spinning disk, LSM, etc.).
